O-16: Metabolism of Exogenous Fatty Acids, Fatty Acid-Mediated Cholesterol Efflux, PKA and PKC Pathways in Boar Sperm Acrosome Reaction

Background: For understanding the roles of fatty acids on the induction of acrosome reaction which occurs under association of cholesterol efflux and PKA or PKC pathways in boar spermatozoa, metabolic fate of alone and combined radiolabeled 14C-oleic acid and 3H-linoleic acid incorporated in the sperm was compared, and behavior of cholesterol and effects of PKA and PKC inhibitors upon fatty aci...

Incorporation of fatty acids containing photosensitive groups into phospholipids of Escherichia coli.

A series of new fatty acids containing photosensitive groups at different positions on the paraffin chains supported the growth of an auxotroph of E. coli requiring unsaturated fatty acids. The derivatives were 6-, 9-, 11-, and 12-azidostearic acids, 12-azido-oleic acid, 16-azidopalmitelaidic acid, and 12-(4-azido-2-nitrophenoxy)-stearic and -oleic acids. Analyses of the phospholipids from cult...

Fluid membranes with acidic domains activate DnaA, the initiator protein of replication in Escherichia coli.

Acidic phospholipids in a fluid phase dissociate ADP or ATP tightly bound to DnaA protein and, in the presence of ATP and DNA, can restore an inactive ADP form to full activity (Sekimizu, K., and Kornberg, A. (1988) J. Biol. Chem. 263, 7131-7135). Further studies of the interactions between DnaA protein and lipids have used two functional assays: 1) release of ADP or ATP from DnaA and 2) DNA re...

Inhibition of calcium-independent phospholipase A2 prevents arachidonic acid incorporation and phospholipid remodeling in P388D1 macrophages.

Cellular levels of free arachidonic acid (AA) are controlled by a deacylation/reacylation cycle whereby the fatty acid is liberated by phospholipases and reincorporated by acyltransferases. We have found that the esterification of AA into membrane phospholipids is a Ca(2+)-independent process and that it is blocked up to 60-70% by a bromoenollactone (BEL) that is a selective inhibitor of a newl...

Incorporation of Photosensitive Fatty Acids into Phospholipids

In an approach to the study of phospholipid-protein interactions in biological membranes, the photoactivable fatty acids, w-(m-azidophenoxy)-undecanoic acid (I) and o-(m-diazirinophenoxy)-hexadecanoic acid (11), were incorporated biosynthetically into the phospholipids of the Escherichia coli fatty acid auxotroph, strain K1060-B5. The extent of incorportation of the two fatty acids was 43% and ...
